Cattle Semen Cold Chain in Bangladesh: Liquid Nitrogen Tanks, Dry Shippers and Handling Rules

Artificial insemination has become an important tool in Bangladesh’s dairy and beef sectors, helping farmers improve the genetics of their herds without keeping breeding bulls. Behind every insemination is one of the most demanding cold chains in agriculture. Frozen bull semen is stored in thin plastic straws, kept in liquid nitrogen at around minus 196 degrees Celsius, and must stay at that temperature from the moment it is frozen at the production centre until the technician thaws it minutes before use. A single lapse, even a brief one, can reduce fertility without any visible sign.

This guide explains how the frozen semen cold chain works, the equipment involved, the handling rules that protect quality, and what importers and service providers in Bangladesh should consider when bringing semen and liquid nitrogen equipment into the country.

Why semen needs liquid nitrogen, not an ordinary freezer

Ordinary freezers, even very cold ones, are not cold enough to stop the slow biological changes that damage sperm cells over time. At the temperature of liquid nitrogen, those processes are effectively stopped, which is why properly frozen semen can be stored for long periods. The challenge is that sperm cells are also very sensitive to temperature fluctuations. If a straw warms up partly and is then returned to the nitrogen, ice crystals inside the cells can change and damage them. The straw looks exactly the same afterwards, but its fertility may be lower.

That is the core idea behind every handling rule in this cold chain: the straw must never be allowed to warm, even briefly, until the moment it is deliberately thawed for use.

The equipment in the chain

  • Storage tanks (cryogenic dewars) — vacuum-insulated containers holding liquid nitrogen. Inside, straws are kept in goblets held in canisters that hang from the neck of the tank.
  • Field or transport tanks — smaller dewars used by AI technicians to carry semen to farms.
  • Dry shippers — specially designed containers in which liquid nitrogen is absorbed into a porous material, so there is no free liquid to spill. They are commonly used for transport, including by air, because they maintain the required temperature without loose liquid.
  • Liquid nitrogen supply containers for refilling the tanks.
  • Thawing units — temperature-controlled water baths used to thaw straws correctly before insemination.
  • Measuring tools — a dipstick or scale to check nitrogen level, since tanks lose nitrogen gradually through evaporation.

Handling rules that protect fertility

Most quality losses in the semen cold chain happen during routine handling, not during long storage. Widely taught good practice includes:

  • Keep the canister low. When locating a straw, raise the canister only as far as necessary and keep it below the neck of the tank, where the cold vapour protects it. Work quickly.
  • Use tweezers or forceps to remove straws, never fingers, which warm the straw immediately.
  • Know where each bull’s semen is before opening the tank, using an inventory record, so straws are not lifted out repeatedly while searching.
  • Never return a straw to the tank once it has been lifted out of the cold zone for too long.
  • Check nitrogen level regularly and refill well before it runs low. A tank that runs dry will destroy its entire contents.
  • Thaw according to the producer’s instructions, at the recommended water temperature and time, and use the semen promptly after thawing.

Tank care and monitoring

A cryogenic tank is a vacuum flask, and its insulation depends on the vacuum between the inner and outer walls. Dropping a tank, dragging it across a floor, or banging it during transport can damage the vacuum. A damaged tank loses nitrogen much faster than normal, sometimes with frost forming on the outside as a warning sign. Tanks should be stored upright, in a clean, dry, ventilated place, off bare concrete, and transported secured so they cannot tip.

Safety: liquid nitrogen is hazardous to people

Liquid nitrogen is not toxic, but it is dangerous in two ways. First, it causes severe cold burns on contact with skin or eyes, so gloves and face protection should be used when filling tanks. Second, as it evaporates it releases large volumes of nitrogen gas, which displaces oxygen. In a small, closed room, a vehicle cabin or an unventilated store, this can create an oxygen-deficient atmosphere without any smell or warning. Tanks should be stored and refilled in ventilated areas, and never carried inside closed passenger cabins. Sealed containers must never be used for liquid nitrogen, because pressure builds up as it boils.

Import and transport considerations in Bangladesh

Frozen semen is a biological product and its import is regulated. Importers generally need permission from the relevant livestock authority, health certification from the exporting country, and compliance with any breed, disease and testing conditions that apply. These requirements can change, so confirm the current rules with the Department of Livestock Services before placing an order. Good records make good handling possible. Every tank should have a map showing which canister and goblet holds which bull’s semen, with the number of straws remaining. When a technician knows exactly where to go before opening the tank, the canister is raised once, the straw is taken, and the canister is lowered again within seconds. Without a map, technicians lift canisters repeatedly while searching, which is exactly the kind of exposure that damages fertility.
